Stable Down-Regulation of HLA Class-I by Serum Starvation in Human PBMCs

چکیده مقاله:
Background: The human leukocyte antigen (HLA) matching between organ donor and recipient is an acceptable strategy in clinical transplantation since 1964. However, in bone marrow transplantation, finding matched donors is often problematic. Thus new method for down regulation of HLA can be an alternative strategy to solve this problem. Objective: To examine the effect of serum starvation on HLA class I expression in human peripheral blood mononuclear cells (PBMCs). Methods: PBMCs were cultured in RPMI-1640 supplemented with 10% FBS (non-starved cells) as well as in medium only (starved cells) for 16, 24, 48, 72, 96h under standard cell culture conditions. The pattern of cell death and HLA class I expression was determined by flowcytometry. Antigenicity of the starved PBMCs was evaluated in a one-way mixed lymphocyte culture by MTT assay. Results: Mean fluorescence intensity (MFI) of different indicated starved PBMCs gradually decreased and this reduction was stable after 96h of re-feeding with medium containing FBS. Under serum starvation condition, PBMCs showed apoptotic cell death pattern. There was a linear correlation between percentages of cells, which exhibited the late apoptosis death pattern and serum starvation period (r=0.88, p

متن کاملHuman Papillomavirus and Cancer - Immunological Consequences of MHC Class I Down-Regulation
Few studies have been conducted on the causative factors associated with the development of cancer. Infection by high risk human papillomaviruses (HPVs) have been implicated as causative agents in a variety of cancers. HPV is capable of evading immune system and establishing persistent infections. Prolonged infection and lesion maintenance are associated with higher risk of neoplastic progressi...

متن کاملHLA class I antigen down-regulation in primary ovary carcinoma lesions: association with disease stage.
PURPOSE To investigate TAP1, TAP2, and HLA class I antigen expression in primary ovarian carcinoma lesions and to assess the clinical significance of defects in the expression of these molecules. EXPERIMENTAL DESIGN Fifty-one formalin-fixed, paraffin-embedded primary ovarian carcinoma lesions were stained with affinity-purified rabbit anti-TAP1 and anti-TAP2 antibodies and with anti-HLA class...
متن کاملHLA CLASS I IN IRANIAN P SORIATIC PATIENTS
The association of HLA and disease varies in different populations. We have . studied the association of HLA class I in Iranian psoriatic patients, in order to compare this association with other reports. Fifty-one Iranian patients with psoriasis were HLA typed. The frequency of HLA antigens in patients and patient subgroups, based on clinical patterns, age of onset, family history and prov...
متن کاملHLA Class I and Class II Genes Distribution of the Sistanis in Iran
Background: The high polymorphism in the human leukocyte antigen (HLA) genes can be used as an identity of individuals to compare with other populations. This extreme polymorphism in the HLA system is accountable for the differences in alleles and haplotypes among ethnic groups, populations, and the inhabitants of many regions. Objective: To define the frequency of HLA alleles and haplotypes am...
